Biofilm Director-You ‘ll die watching me

Biofilms consists of microbial sessile communities characterized by cells that colonise surfaces embedded in a matrix of extracellular polymeric substances .Some of major food pathogens are biofilm formers such as Eischerichia coli, Campylobacter jejuni causing a major food safety challenge. So the aim of this research is to isolate and determine best mitigation strategies of biofilm formers on farm fresh produce.

Take 1
Hello everyone . I am a biofilm director ,many biofilms were directed under me and gaining celebrity status.Recently in centre of food science and technology, CCSHAU, Hisar,I stood by for take 1.A researcher bought fresh baby carrot, cabbage and spinach and by prioviding extreme conditions, I decided to grow a few characters for my biofilm . There were many bacterial colonies growing in media I.e. their food to nourish their hair(flagella)to showcase who is more sessile.So then after 2-3 months of determined researcher I supported the researcher by screening the biofilm formers.The black colored colonies on a dye known as Congo red and BHIA(Brain heart infusion agar) were decided as my lead players.They were hot and sexy to be considered as isolates whom I could groom and then help them play their part in the final biofilm making.


Take 2
Now we had 7 isolates which had to characterise to know which part had to be played by whom so here comes the role of 16SrDNA gene amplification through PCR.
These genomic DNA were now having multiple copies of their ownself.We now send their copies for sequencing so that we could give name to their characters whether Staphylococcus, Streptococcus or

Bacillus.
Also their sequencing was necessary to know which sequel of the film will be majorly taken by which isolate .

Take 3
Till the time their portfolio was sent to outsourced places, we have totest them in other ways like their biofilm assay efficacy which will ensure their strength to keep working for this film for how long they will maintain their junction with polystyrene tubes.
aLso, their hydrolytic activity for various enzymes need to be checked.
The enzymes will empower them to fight for their growth in host body and let them stay and enjoy human gut .Human gut is the place where whole film will be recoreded.

Take 4
Now we will identify their role as food pathogens,.The most leading character will have serial killer role who may kill infants, lactating mothers, immuno compromised individuals and old hearts.
Well, serial killer sounds like a thriller biofilm movie. I will have lots of fan on Microgram,Hemocytogram and SEM where easy biopics can also be made. I Just love my work
Take 5
Now I have to know how these isolates react when pigments are added, beta carotene sounds to be a great fan of those obtained from carrot , Hey!ya cabbabge is good with exopolysaccharides too so its total sugars are just candy for all the sweet toothed isolates.
Acidity and pH yes, the power of hydrogen will tell their power to grow in a range of 1-14
Oops!they died in tryptic soy broth which was nearly stale.
Have to know their moisture content so that they don’t easily die out when served food in plates named PETRI haha ..funny naa!

Take 6
Thank God, they didn’t die out of low water activity ,. Yes they are active in water which is present from outside environment.O I can see something, they have got other talents like maintaining active life even when runniong short of water because they stay in B.O.D. incubator that supports their oxygen level.wow!they have their personal trees in lab.

Take 7
Its time to eliminate them one by one as we need a lead player for our bio-FILM.
They will be eliminated using UV light, chemical prservatives, salt,various temperature conditions etc.
Nisin can also be used for eliminating them in ppm
In the meanwhile they were all showcasing blue rimgs within a tube which indicated them to be super fit for their role as BIOFILM formers.A purple colred dye was used to get rings in a trypic soy broth medium where they were allowed to grow to showcase their individuyal talents alongside juries Staphylococcus aureus, E. coli, Bacillus subtilis




And they were all best competitors to each other.2 of them formed better purple rings in the middle of the test tubes….yaa the proven heroes.

Take 8
Now its time for semi finals
They were given treatments of various parameters and they hand;led all well.
Some of them died at the hands of temperature , yet other at salt concerntrations and other with the exposure to UV light and other bacteriocins

Take 9
They were observed more for inhibition and treated on spoiled farm fresh produce. First ,they were intentionally spoiled , but because of these treatments these isoltes all forgot their role as jury was damn strict.they were left to denote zone of inhibition. This zone added to their melancholy strain and they started moving to their decilne phase and ultimately died out of shortase of food. Food stress is tough to tolerate for everyone.
The hungerbugs could not survive the hostile environment of external factors thus, leading to suscepotibilty towards ampicilin ,a widely studied antibiotic.stress --aah! Poor food pathogens ..poor lead players of my film . I need to re-cast my players for New BioFilm more of strength ,more of resilience . these ressearchers are getting on my nerves. Actually jealous of my fame!

I love humans but am afraid of these researchers…
Hope they lessen the sanctity of their research and work less on disease control.

Well that was fun, a microbiology class disguised as a story. I was suprised to learn that the techniques I learned over fifty years ago for plating things out on agar (the blood one you cite, we used to call "chocolate agar" for capturing the elusive Niesseria Sicca) are still being used. Most of the tracking nowadays I thought was done by dna sequencing.

Yep, water, salt, UV light, all provide ways to alter the environement for either good...or bad. A lot of good Science, and flowcharting of species in this story.
